Research/Clinical Interests

The primary focus of my research is to examine the effects of coagulation proteins, proteases, and receptors in the pathogenesis cardiovascular disease (CVD), specifically atherosclerosis and abdominal aortic aneurysms (AAAs). We utilize genetically modified mice and mouse models of disease to generate data and attempt to verify these results in the human condition, when possible, with retrospective clinical data or human specimens. While several drug therapies exist for atherosclerosis, there are currently no pharmaceutical therapies for patients with AAA. Further, even with modern pharmaceutical regiments for patients with coronary artery disease, it is still the leading cause of mortality. Therefore, the long-term goals of my work are to increase the understanding of thrombosis in atherosclerotic and aneurysmal disease and translate these findings into effective therapeutics to improve survival and quality of life for CVD patients. Atherosclerotic disease is the leading cause of worldwide mortality with an estimated 7.4 million attributable deaths occurring in 2012. Abdominal aortic aneurysm (AAA) affects 5-10% of the male and female population over the age of 65 and is the 10th leading cause of death in the United States. Histopathology of these two arterial diseases is similar with infiltrates of lipids, platelets, vascular smooth muscle cells (VSMC), macrophages (foam cells), T and B lymphocytes, and extracellular matrix resulting in severe inflammation. In atherosclerosis, cell and matrix deposition results in narrowed blood vessels and restricted blood flow leading to thrombus, ischemic stroke, or coronary occlusion; in AAA, such deposition results in a permanent localized dilatation in the arterial wall, formation of intramural clots, elastin and collagen destruction, and life-threatening aortic wall rupture. The fundamental connection between these two arterial diseases is the acute immune reaction to endogenous ligands, i.e. ‘sterile inflammation,’ and activation of the coagulation cascade.
